摘    要:利用规模化生产工艺提高竹荪菌丝体胞内多糖得率,通过对竹荪菌丝体液态深层发酵培养基组分的筛选,采用单因素试验与Box-Behnken响应面法确定竹荪菌丝体液态深层发酵的最佳培养基配方为:葡萄糖32.3 g/L,酵母粉4.5 g/L,磷酸二氢钾2.1 g/L,硫酸镁2.5 g/L。在此条件下竹荪胞内多糖得率的预测值为1.23 g/L,摇瓶实验验证胞内多糖得率为1.19 g/L,与预测值相当。在5、30 L和50 L发酵罐中验证胞内多糖得率分别达到1.43、1.28 g/L和1.26 g/L,分别高于预测值16.26%、4.06%和2.43%。本研究优化获得的竹荪菌丝体液态深层发酵工艺有效促进了竹荪胞内多糖的高产,该工艺可为竹荪多糖的规模化生产应用提供较好的参考。

Abstract:The main purpose of this study was to increase the yield of mycelial polysaccharides from Dictyophora indusiata using a large-scale production process.Using a combination of one-factor-at-a-time method and response surface methodology with Box-Behnken design,the optimum medium formulation for the submerged fermentation of D.indusiata was determined as follows:glucose 32.3 g/L,yeast powder 4.5 g/L,potassium dihydrogen phosphate 2.1 g/L,and magnesium sulfate 2.5 g/L.The predicted yield of intracellular polysaccharides was 1.23 g/L,which was close to the experimental value from shake flask fermentation(1.19 g/L).The yields of intracellular polysaccharides in 5-,30-and 50-L fermentors were 1.43,1.28 and 1.26 g/L,respectively,which were 16.26%,4.06%and 2.43%higher than the predicted values,respectively.In this study,the optimized fermentation process gave a high yield of intracellular polysaccharides.Thus,this process provides a good basis for the large-scale production of D.indusiata mycelial polysaccharides.
