响应面法优化超声波辅助提取黑小麦花色苷的工艺研究

摘    要:以黑小麦为研究对象,利用响应面分析法对黑小麦超声波辅助乙醇提取花色苷工艺进行优化。在单因素试验的基础上,以料液比、乙醇浓度、超声时间、超声温度为影响因素,采用Box-Behnken响应面法,利用四因素三水平正交试验确定最佳提取条件。结果表明,对花色苷提取量影响最大的是料液比,其次是超声温度和超声时间,影响最小的是乙醇浓度。最终确定黑粒小麦的花色苷最佳提取条件是料液比1∶48(g/mL),乙醇浓度60%,提取时间58 min,提取温度40℃。在此条件下,花色苷提取量最高为28.61 mg/100 g,与模型理论预测值相近,说明该模型回归性良好,试验的拟合程度高,可以用于黑粒小麦花色苷提取率的预测。

Abstract:Using black wheat as the research object,and the extraction process of black wheat was optimized by response surface methodology.Based on the single factor test and the initial extraction conditions of anthocyanin,the material-liquid ratio,ethanol concentration,ultrasonic time and ultrasonic temperature were used as the influencing factors,and the Box-Behnken response surface method was used to design 4 factors and 3 levels to determine the best extraction conditions.The results showed that the most important effect on anthocyanin content was the material-liquid ratio,followed by ultrasonic temperature and ultrasonic time.The least affected was ethanol concentration.The optimal extraction conditions for the anthocyanins in black wheat was 1∶48(g/mL),ethanol concentration was 60%,extraction time was 58 min,extraction temperature was 40 ℃. Under this condition, the highest anthocyanin content was 28.61 mg/100 g, which was similar to the theoretical prediction of the model, indicating that the model has good regression and high degree of fitting. It could be used to predict the extraction rate of anthocyanins in black wheat.
