实时荧光定量PCR仪中荧光检测装置的设计

摘    要:设计了基于LED光源的积分球式聚合酶链式反应(PCR)荧光检测装置。系统采用单色LED作为光源,经过积分球的均匀辐射后,减少光源对样品的影响,再通过光电倍增管与后续电路,得到PCR循环中的实时荧光光强,这有效地减小了荧光检测装置的体积,提高了仪器的性价比。实验通过装置和iCycler-iQ5实时荧光PCR仪的检测数据进行比对,仪器偏差分别为9.44%和25.98%,发现装置具有更高的精确度,更好的稳定性和重复性。

Abstract:PCR fluorescence detection device is designed based on LED and integrating sphere.System used monochromatic LED as the light source,which through the integrating sphere of uniform radiation to reduce the influence of light source on the sample.Through the photomultiplier tube and follow-up circuit,the real-time PCR cycle fluorescence intensity is gotten.This design could effectively reduced the volume of fluorescence detection device and improved the cost-effective of instrument.This experimental results is compared with the results of iCycler-iQ5 PCR instrument.The instrumental deviations are 9.44 % and 25.98 % respectively.It shows that this device has higher precision,better stability and reproducibility.
