| 摘 要： | 目的对临床分离的5株鸡源肺炎克雷伯菌进行菌毛类型的鉴定。方法首先利用传统的D-甘露糖血凝特性(MSHA/MRHA)试验对临床分离的5株鸡源肺炎克雷伯菌进行菌毛分型,同时以该菌Ⅰ型和Ⅲ型菌毛的主要结构亚单位(fimA/mrkA)基因序列为靶序列,分别设计一对引物,利用PCR扩增的方法对该5株菌进行菌毛分型。结果MSHA/MRHA方法鉴定其中的4株菌具有Ⅲ型菌毛,另外1株无法鉴别,而PCR方法鉴定5株菌均具有Ⅲ型菌毛。结论该5株鸡源肺炎克雷伯菌均具有Ⅲ型菌毛。MSHA/MRHA具有方便、快速的特点,但不敏感;PCR则具有快速、敏感、准确的优点。研究结果对于肺炎克雷伯菌的流行病学调查及该菌的临床诊断具有指导意义。

| Abstract: | Objective To identify the types of fimbriae of 5 clinical isolates of avian Klebsiella pneumoniae.Methods Type the fimbriae of 5 isolates by MSHA/MRHA.Meanwhile,design a pair of primers according the gene sequence of main structural subunits(fimA/mrkA)of fimbriae types Ⅰ and Ⅲ of avian Klebsiella pneumoniae respectively and identify the types of fimbriae of the 5 isolates by PCR.Results By MSHA/MRHA,the fimbriae of 4 of the 5 isolates were identified as type Ⅲ,and the rest one as undefined type.However,by PCR,all the fimbriae of 5 isolates were identified as type Ⅲ.Conclusion All the 5 isolates possess fimbriae type Ⅲ.MSHA/MRHA was simple and rapid but insensitive for typing of fimbriae.However,PCR was rapid,sensitive and accurate.The study is of guiding significance in epidemiological investigation and clinical diagnosis of infection with avian Klebsiella pneumoniae. |
