摘    要:利用筛选的肠杆菌发酵生产壳聚糖酶,对培养温度、pH和接种量进行优化,确立中心位点后进行响应面设计分析,确定最优发酵培养条件。结果表明:最佳培养工艺条件为培养温度31.74℃、pH6.5和接种量为7.26%(V/V),培养36h时壳聚糖酶活性达到13.59U/mL,实际值与预测值之间的误差约为0.43%。壳聚糖酶的生产方式为延续合成型,具有深入优化并规模生产的潜力。

Abstract:Optimization of culture conditions were conducted for chitosanase production by Enterobacter sp. YT21. Firstly Culture temperature, pH and inoculum volume were optimized by one-factor-at-a-time experiments, and then Box-Behnken design and response surface methodology were employed for further optimization of the chitosanase production. The results indicated that the optimal culture conditions were 31.74℃, pH 6.5 and 7.26% (V/V) inoculum, and the highest chitosanase activity obtained was 13.59 U/mL. Error of the actual value compared with the predicted one was only 0.43% , so Enterobacter sp. YT21 was proved to be a suitable potential microorganism for use in chitosanase production.
