摘    要:小分子质量右旋糖酐在医学、化工、食品等领域有广泛的应用,为了得到特定分子质量的右旋糖酐,采用酶解法对大分子质量右旋糖酐的分子质量及分子质量分布进行调控.实验结果表明:在酶解过程,通过控制右旋糖酐酶浓度、底物浓度和反应温度、pH值及时间,可有效调控右旋糖酐的分子质量,且产物均一性较好.较适宜酶解条件:酶浓度为10 U/m...

Abstract:Small molecular weight dextran has been widely used in medicine,chemical industry,food and other fields.In order to obtain specific molecular weight dextran,enzymatic hydrolysis was used to control the molecular weight and molecular weight distribution of high molecular weight dextran.The results show that the molecular weight of dextran can be effectively controlled by controlling the concentration of dextranase,substrate,reaction temperature,pH and time,and the product homogeneity is good.The optimum conditions were as follows:enzyme concentration 10 U/mL,reaction temperature 50 ℃,pH 5.0,substrate concentration 30 mg/mL.The structure of dextran did not change before and after enzymatic hydrolysis,but its conformation changed from spherical chain to compliant chain.When the molecular weight of dextran was about 5000 u,it was a compact spherical chain conformation in solution.Enzymatic degradation of dextran is fast,mild reaction conditions,low energy consumption and pollution-free,so it has a good application prospect in the production of dextran with specific molecular weight.
