Abstract:A method using solid phase extraction-high performance liquid chromatography with diode array detection (SPE-HPLC-DAD) was developed for the determination of bongkrekic acid in starch and its products.The sample were extraction with ammonia methanol and cleaned up with P-SAX solid phase extraction column.The target compound was separated on a C18 column (4.6 mm×250 mm,5 μm),using methanol-0.1% formic acid water (80:20,v/v) as the mobile phase.The quantification analysis was performed by the external standard method with the wavelength of 271 nm.The results indicated that an excellent linear relationship in the range of 0.02~20 μg/mL (R2=0.998).The recoveries of bongkrekic acid in starch and its products at three spiked levels ranged from 96.5% to 105.1% with a relative standard deviation less than 5%.The method is easy to operate and accurate,which is of great significance for the quality and safety control of starch and its products.
