产菊粉酶菌株的筛选及发酵条件研究

摘    要:自山东威海沿海滩涂菊芋生长根际土壤中,筛选得到具有菊粉酶能力的菌株25株,通过复筛得到1株具有高产菊粉酶活力的真菌,通过形态学观察、18S rDNA全序列分析,初步鉴定该菌为囊状青霉(Eladia saccula SA1201)。对其发酵产酶条件的研究结果表明,最适产酶发酵条件为菊粉30.0g/L,蛋白胨7.0g/L,NaCl 5.0g/L,K2HPO4.3H2O 3.0g/L,初始pH值6.0,250mL发酵三角瓶装液量为50mL,30℃、160r/min振荡培养3d。

Abstract:A strain with high inulinase productivity was screened, and identified as Eladia saccula SA1201 through observation by morphologic and identification of 18S rRNA gene partical sequence. The fermentation conditions of the strain were optimized by orthogonal test. the optimal culture medium consisted of 30.0g/L inulin, 7.0g/L peptone, 5.0g/L NaC1, 3.0g/L K:HPO4·3H2O, initial pH value 6.0, medium volum 50ml/250ml, 30℃ and 160r/min for 3d.
