摘    要:建立了苦杏仁中苦杏仁苷的高效液相色谱-蒸发光散射检测器检测方法。采用ZORBAX Eclipse XDB-C18色谱柱(4.6mm×150mm,5μm),流动相为V(甲醇)∶V(水)=40∶60,流速1.0mL/min,柱温40℃;蒸发光散射检测器漂移管温度60℃,载气(空气)压力0.2MPa。在上述条件下测得苦杏仁苷在0.28~5.5μg范围内的线性关系良好(r=0.9975),检出底限达58ng/L,平均加标回收率为96.2%。该方法简便快捷,重现性和准确度良好,可用于实际工作中样品的测定。

Abstract:A high performance liquid chromatographic method with evaporative light scattering detector(ELSD)was established for determination of Amygdalin in Bitter Almond.Amygdalin was successfully separated by a XDB-C18 column(4.6x150mm,5um)at 40℃,as mobile phase composition with a binary solvent mixture of methanol-water(40∶60,volume ratio).The flow rate was 1.0mL/min.The ELSD drift tube temperature was 60oC, carrier air pressure was 0.2MPa.In this condition the linear range was 0.28~5.5μg(r=0.997 5).The detection limit was 58ng and the average recovery was 96.2%(RSD=2.6%,n=4).This method was accurate,rapid,simple,and reproducible。It is suitable for the sample detection in practice.
