Process Analysis to Target Novel CO2-Expanded Ethanol Extraction of Grape Seed Oil by Visualization through Microscopic Imaging
Abstract:Visualization technology by microscopically imaging was developed aiming to unveil the mechanism of extracting linoleic acid-rich grape seed oil with a novel pressurized solvent, CO2-Expanded Ethanol (CXE). Firstly, the multi-microcavity structure of seed particle matrix was revealed through imaging of Scanning Electron Microscopy (SEM). By mean of specific oil-staining with Nile Red dye, fluorescent quantitative method had been established to precisely investigate the three-stage process of seed oil extraction under the high operation pressure. Moreover, Confocal Laser Scanning Microscopy (CLSM) was used to further exactly visualize the spatial distribution of seed oil inside the matrix particle, so as to intuitively analyze the oil release from inside to outside. Finally, the new CXE extraction was evaluated from the microscopic scale, compared with the other traditional methods. In conclusion, establishment of microscopic visualization methods is helpful to intuitively learn the crucial information such as the orientation of oil as well as mass transfer during the extraction, illuminating the mechanism of complex solvent involved extraction. Thus, it would not only give the valuable evidences for classic theoretical analysis, but also boost extensive application of new techniques.
