立体选择性还原芳香酮微生物的筛选及反应特性的研究

摘    要:以苯乙酮为模型底物,从土壤中筛选得到了两株能高效催化前手性芳香酮不对称还原分别生产相应的S-和R-型醇的菌株,鉴定后分别为白地霉(Geotrichum candidum)和毕氏酵母(Pichia pastoris)。进一步考察了这两株菌催化苯乙酮不对称还原的反应特性,发现毕氏酵母还原苯乙酮的产物以R-型(R-α-苯乙醇)为主,产率达到75%左右,e.e.达到90%左右;白地霉还原苯乙酮的产物为S-α-苯乙醇为主,产率达到80%左右,e.e.达到70%左右。这较面包酵母具有更高的催化活性。

Abstract:The microbes for asymmetric reduction of prochiral aromatic ketones were isolated from soil using acetophenone as the sole carbon source,when asymmetric reduction of acetophenone(ACP) to chiral α-phenethyl alcohol(PEA) was chosen as the model reaction.Two microbe strains with excellent catalytic activity were obtained.They were Geotrichum candidum and Pichia pastoris by bacteria identification.The product of asymmetric reducuion of ACP by Pichia pastoris was mainly R-PEA and that by Geotrichum candidum was mainly S-PEA.The yield and e.e.could respectively reach 75% and 90% for Pichia pastoris,and 80% and 70% for Geotrichum candidum,much higher than those catalyzed with baker′s yeast.
