摘    要:目的建立以胶原蛋白-肾上腺素诱发小鼠体内血栓形成与角叉菜胶诱发大鼠尾部血栓形成模型,评价重组瑞替普酶(Reteplase)对体内诱发血栓形成的快速溶栓作用,为临床应用提供依据。方法将Reteplase与诱导剂混匀后给小鼠作尾静脉注射,观察给药后5min内小鼠死亡数和15min内小鼠偏瘫恢复数,计算药物对小鼠形成脑血栓的保护率;大鼠足跖皮下注射角叉菜胶使尾部形成血栓后给药,观察血栓平均长度及再通率。结果Reteplase对胶原蛋白-肾上腺素诱发小鼠体内血栓有迅速溶栓、急救效应;对角叉菜胶诱发大鼠尾部混合血栓有显著溶栓作用。结论溶栓作用呈现明显的剂量-效应关系。本文首次报道了Reteplase对体内诱发血栓的溶栓作用。

Abstract:Objective To establish the thrombosis model in mice induced by collagen-adrenalin in vivo and the rat mode induced by carrageenin and to evaluate the rapid thrombolysis of recombined Reteplate to blood clots induced in vivo so as to guideline its clinical applications. Methods Reteplase mixed with inductors was administrated to mice by tail intravenous injection. The number of death mice in five minutes after administration and number of hemiple- gia recovery in fifteen minutes were both observed for accounting the protection rate. Rats induced by voix pedis subcutaneous injection with carrageenin for thrombose were also administrated for the observation of thrombus average length and recanalization rate. Results The Reteplase could rapidly dissolve the blood clots in mice induced by collagen-adrenalin and show emergence treatment to mice. Also it is effective to the composed blood clots induced by carrageen in rats. Conclusion The remarkably thrombolysis displays the dosage-effect relationship. This article originally reports reteplase with the effect of rapid thrombolysis.
