偶氮硝羧与血清蛋白质反应的分光光度研究

摘    要:在pH3.0的BrittonRobinson缓冲介质中,红色的偶氮硝羧与蛋白质形成蓝紫色复合物1],其最大吸收波长约为680nm,比试剂本身红移140nm,表观摩尔吸光系数达2.60×105Lmol·cm(牛血清白蛋白),线性范围为20~140μgmL。实验制定的几种蛋白质的标准工作曲线,其线性范围与牛血清白蛋白一致,灵敏度高。用于人血清样品测定,方法可靠。

Abstract:It was found that red carboxynitroazo can react with serum proteins forming a blue violet complex in Britton Robinson buffer at pH 3.0 and room temperature, which gives maximum absorption peak at 680 nm with 140 nm red shift in from that of carboxynitroazo. A molar absorptivity of 2.60×10 5 L/mol·cm (BSA) and Lineary range of 20~140 μg/mL for proteins were determined. Standard curves were constructed under optimum conditions obtained in our lab. The Lineary range is wide. This assay is sensitive, accurate and is tole rant of many foreign substances. Moreover, the reagents are stable and this method is easy to carry out.
