鳗弧菌pEIB1质粒缺失株MVAV6201的筛选和三价铁源供给的优化

摘    要:以鳗弧菌Vibrio anguiUarum MVM425(01型)为出发菌株,通过变温培养诱变筛选得到pEIB1质粒缺失株MVAV6201。通过与野生株MVM425的摇瓶培养对比发现,Fe^3 营养的摄取是质粒缺失株MVAV6201生长的瓶颈。MVAV6201对Fe^3 的吸收有赖于外界Fe^3 营养的供给形式。柠檬酸铁铵能有效增强MVAV6201对三价铁营养的吸收,促进菌体细胞的生长。柠檬酸铁铵饱和浓度为0.4mmol/L。

Abstract:A plasmid pEIB1 cured strain of Vibrio anguillarum MVAV6201 was attained from the wild MVM425 (Serotype O1) strain by the method of temperature elevated incubation. It was found in the comparative experiments between plasmid cured MVAV6201 and wild MVM425 that the growth of MVAV6201 was limited by the poor ability of iron uptake. The iron uptake of MVAV6201 depended on the way that iron was supplied. Ammonium ferric citrate was found to greatly improve the iron uptake and growth of the plasmid cured cells. The saturated concentration of ammonium ferric citrate for cell growth was 0.4 mmol/L.
