(2S,4S)-4-羟脯氨酸的合成工艺研究

摘    要:以(2S,4R)-4-羟脯氨酸为基础原料,以水为介质,利用三苯基膦和醇的Mitsunobu合成反应,改变其构型得到(2S,4S)-4-羟脯氨酸。此方法在现有合成路线基础上有所改进,在实验过程中探索反应物配比对各步反应收率的影响,得到实验的最优条件。并对产物进行表征及纯度分析,确定为(2S,4S)-4-羟脯氨酸,纯度为87.2%,收率93.3%。

Abstract:Using (2S,4R)-4-hydroxyproline as the raw material,water as the medium,(2S,4S)-4-hydroxyproline was synthesized through Mitsunobu reaction of triphenylphosphine and alcoheol to change the configuration of (2S,4R)-4-hydroxyproline.This method was improved based on the existing synthetic route.The influence of reactant molar ratios on the yield of each reaction step was investigated.The optimal experimental condition was obtained.The product was characterized by 1HNMR and liquid chromatogram and its purity was analyzed.The product was identified as (2S,4S)-4-hydroxyproline,and its purity and yield were 87.2% and 93.3%,respectively.
