啤酒酵母生物合成胞苷三磷酸的实验研究

摘    要:以胞苷一磷酸(CMP)为底物,利用啤酒酵母细胞内的酶系催化合成了胞苷三磷酸(CTP),实验采用高效液相色谱分析了底物浓度、葡萄糖浓度、磷酸盐浓度、酵母量、反应时间等因素对转化反应的影响。结果表明,最佳转化条件为:CMP 20 mmol/L、葡萄糖150 mmol/L、磷酸盐250 mmol/L、酵母量0.3 g/mL、反应时间75 min。在此条件下,CTP的平均产率可高达59.01%。

Abstract:Cytidine triphosphate(CTP)produced from cytidine monophosphate(CMP)utilizing enzymes in saccharomyces cerevisiae was studied. The effects of some key factors,such as the concentration of CMP,glucose,phosphate,yeast cells and reaction time on the biotransformation process were studied by HPLC method. Results showed that the optimal conditions were obtained as follow:20 mmol/ L CMP,150 mmol/ L glucose,250 mmol/ L phosphate,0. 3 g/ mL yeast and reaction time 75 min. Under optimal conditions,yield of CTP was up to 59. 01% .
