正己烷-乙醇-水三元双相溶剂浸提法富集南极磷虾磷脂

摘    要:为研究利用正己烷-乙醇-水三元双相溶剂浸提法从南极磷虾中提取富集磷脂,采用乙醇提取南极磷虾的总脂质,以总脂质得率为评价指标,考察料液比、浸提时间、浸提温度、浸提次数对总脂质提取效果的影响;而后以脂质得率、磷脂含量、磷脂得率为评价指标,优化采用正己烷-乙醇-水三元双相溶剂从南极磷虾总脂质中萃取富集磷脂的最佳条件。结果表明,采用乙醇提取南极磷虾总脂质的最佳条件为料液比1∶3(g/mL)、浸提时间3 h、浸提温度40 ℃、浸提3 次,在该条件下,南极磷虾总脂质得率为(4.69±0.13)%。正己烷-乙醇-水三元双相溶剂从南极磷虾总脂质中萃取富集磷脂的最佳体积比为0.275∶0.600∶0.125,获得的南极磷虾磷脂中磷脂含量为(85.46±0.31)%。南极磷虾磷脂主要为磷脂酰胆碱,磷脂中二十碳五烯酸(eicosapentaenoic acid,EPA)和二十二碳六烯酸(docosahexaenoic acid,DHA)的总量达到总脂肪酸含量的(53.39±0.76)%。

Abstract:The extraction and enrichment process of phospholipids from Antarctic krill(Euphausia superba)using two-liquid-phase system of hexane-ethanol-water was investigated in this study. Ethanol was used to extract total lipids from Antarctic krill,and the yield of total lipids was taken as the evaluation index.The effects of material-liquid ratio,extraction time,extraction temperature and extraction times on the extraction efficiency of total lipids from Antarctic krill were firstly evaluated.Subsequently,the optimal condition for enriching phospholipids from the total lipids of Antarctic krill using the two-liquid-phase system of hexane-ethanolwater was optimized with lipid yield,phospholipid content and phospholipid yield as the evaluation indexes.The results showed that the optimal conditions for extracting total lipids from Antarctic krill using ethanol were a material-liquid ratio of 1∶3(g/mL),extraction time of 3 h,extraction temperature of 40 ℃,and extraction 3 times. Under these conditions,the yield of total lipids from Antarctic krill was(4.69 ± 0.13)%. The optimal volume ratio of hexane-ethanol-water for enriching phospholipids from the total lipids of Antarctic krill was 0.275∶0.600∶0.125. The phospholipid content in the obtained Antarctic krill phospholipids reached up to(85.46±0.31)%.The main component of Antarctic krill phospholipids was phosphatidylcholine,and the total amount of eicosapentaenoic acid(EPA)and docosahexaenoic acid(DHA)in phospholipids accounted for(53.39±0.76)% of total fatty acid content.
