肝素黄杆菌菌种保存方法的研究

摘    要:目的研究多种糖类对肝素黄杆菌细胞的保护作用及保藏菌种的方法。方法将多种糖类加入黄杆菌悬液,观察冷冻、冷藏、冷冻干燥过程中肝素黄杆菌的存活率,并将海藻糖和甘露醇添加进斜面固体培养基,观察菌种的有效保藏时间。结果糖类的加入明显提高了肝素黄杆菌菌种保存的存活率,其中以海藻糖和甘露醇效果最佳。结论在培养基中添加2%海藻糖或2%甘露醇可使菌株斜面冷藏保存时间延长2周。

Abstract:Objective To study the protection effect of added saccharides on flavobacterium heparinum cells and to explore the use of saccharides in strain conservation.Methods The survival of the bacteria during the process of freeze-thawing,cold storage or lyophilization was investigated with many kinds of saccharides to be added into the suspension of flavobacterium.The preservation period was assayed with 2% trehalose or 2% mannitol in the slant solid medium.Results The survival of flavobacterium heparinum solutions containing saccharides was much higher than control group,especially with trehalose or mannitol in the medium.Conclusion Trehalose and man-nitol are two of the best protect additives among used and they with 2% concentration alone can prolong the preservation period for two weeks increased.
