高效液相色谱法测定化妆品中7种酚羟基类美白成分

摘    要:建立了高效液相色谱-二极管阵列检测器(HPLC-DAD)同时测定化妆品中熊果苷、氢醌、阿魏酸、苯酚、丁基间苯二酚、苯乙基间苯二酚和己基间苯二酚7种酚羟基类美白成分的方法。用甲醇提取样品,采用Kromasil C18色谱柱(250 mm×4.6 mm×5μm)分离,以甲醇-质量分数0.02%磷酸作为流动相进行梯度洗脱,流速1.0 mL/min,柱温30℃,检测波长280 nm。熊果苷、氢醌、苯酚、丁基间苯二酚、苯乙基间苯二酚、己基间苯二酚的质量浓度在2~200 mg/L,阿魏酸的质量浓度在0.5~50 mg/L范围内,线性关系良好,相关系数均为0.9999,且保留时间和峰面积的日内与日间精密度均良好,相对标准偏差(RSD)小于2.0%。此方法检出质量比为1~4 mg/kg,回收率为96.2%~112.3%,相对标准偏差(RSD)为0.8%~2.1%。该方法简单、便捷、快速、准确,可用于化妆品中美白成分的测定。

Abstract:A method based on high performance liquid chromatography-diode array detector(HPLC-DAD)for simultaneous determination of arbutin, hydroquinone, ferulic acid, phenol, butyl resorcinol, phenylethyl resorcinol and hexyl resorcinol in whitening cosmetics was established.These seven compounds were extracted by methanol. Kromasil C18 column(250 mm × 4.6 mm × 5 μm) was used with mobile phase of methanol-mass ratio of 0.02% phosphoric acid for conducting gradient elution.The flow rate was 1.0 mL/min, column temperature was 30 ℃ and detection wavelength was 280 nm. Under this condition, there were good linear relationships between the mass concentration and the peak area for arbutin, hydroquinone, phenol, butyl resorcinol,phenylethyl resorcinol and hexyl resorcinol in the range of 2-200 mg/L, for ferulic acid in the range of 0.5-50 mg/L, respectively. All of their correlation coefficients were 0.9999. And the intra and inter day precision of retention time and peak area were also good with relative standard deviations(RSD) less than 2.0%. The limits of detection were 1-4 mg/kg. The average recovery rates were from 96.2% to 112.3%,and the relative standard deviations(RSD) were from 0.8%to 2.1%. This method is simple, convenient,accurate,reliable and suitable for the determination of seven compounds at the same time.
