摘    要:该研究建立了一种利用高效液相色谱法测定黑曲霉(Aspergillus niger)xj发酵液中5-羟甲基糠醛(5-HMF)含量的检测方法。样品经乙醚提取,采用Spheriorb C18柱(4.6 mm×150 mm,5 μm)为色谱柱,以乙睛∶0.1%乙酸水(冰乙酸调)=10∶90(V/V)为流动相,柱温30 ℃;流速1.0 mL/min,检测波长为l=277 nm。在该色谱条件下,5-HMF呈现较好的分离效果和重现性,其含量与峰面积呈现良好线性关系(R为0.999),保留时间2.436 min,回收率87.12%~90.78%,精密度实验结果相对标准偏差(RSD)0.71%。测得发酵液样品中5-HMF含量为0.125 7 mg/g。该方法准确可靠,重复性好,可对黑曲霉发酵液中5-HMF的含量测定作参考作用。

Abstract:In this study, a method for determination of 5-hydroxymethyl-furfural (5-HMF) in fermentation broth of Aspergillus niger xj was established by HPLC. The samples were extracted with diethyl ether, with Spheriorb C18 (4.6 mm×150 mm, 5 μm) as chromatographic column, with acetonitrile and 0.1% acetic acid water (ice acetic acid) = 10∶90 (V/V) as mobile phase at 30 ℃. The flow rate was 1.0 ml/min and the determine wavelength was 277 nm. Under the chromatographic conditions, the 5-HMF showed good separation effect and reproducibility, and its content was in good linear relationship with the peak area (R=0.999). The retention time of 5-HMF was 2.436 min and the recovery rate was 87.12%-90.78%, with relative standard deviation (RSD) 0.71% of precision experiments results. The content of 5-HMF in fermentation broth sample was 0.125 7 mg/g. The method was accurate, reliable and with good reproducibility, and could be used as a reference for the determination of 5-HMF in fermentation broth of A. niger.
