摘    要:蝙蝠蛾拟青霉(Paecilomyces hepiali)是分离自冬虫夏草(Ophiocordyceps sinensis)的一种无性型菌株,其发酵产物具有良好 的生物活性。 该研究利用蝙蝠蛾拟青霉在发酵培养基中液态发酵24 h后产生的无性孢子作为下一批次的发酵接种物,进行了20个批 次的连续循环发酵,减少了发酵种子制备步骤。 各批次发酵的生物量为(25.5~27.2 g/L),孢子数为(0.8×108~1.4×108 spores/mL),腺 苷产量为(49.5~62.3 mg/L),总三萜产量为(126.3~142.7 mg/L),发酵稳定性良好,没有统计学显著差异(P>0.05)。 基于无性繁殖过 程的蝙蝠蛾拟青霉循环发酵方法可以免去各批次菌种斜面培养和种子发酵制备、节约生产成本、提高生产效率。

Abstract:Paecilomyces hepiali was an asexual strain isolated from Ophiocordyceps sinensis, and its fermentation products had good bioactivity. The asexual spores produced by P. hepiali with liquid-state fermentation for 24 h were used as the inoculation of the next batch fermentation. A twenty-cycle reproduction-based repeated batch fermentation (RBF) process of P. hepiali was carried out, and the preparation process of fermented seed was reduced. The biomass, sporulation and bioactive components (adenosine and crude triterpenoids) of each fermentation cycle was determined, and biomass (25.5- 27.2) g/L, spore numbers (0.8×108-1.4×108) spores/ml, adenosine production (49.5-62.3) mg/L, and crude triterpenoids production (126.3-142.7) mg/L showed good batch-to-batch fermentation stability throughout the fermentation process (P>0.05). The asexual reproduction-based RBF process of P. hepiali notably reduced cost and increased production efficiency since omitting out each batch of slant culture and seed fermentation each time.
