摘    要:利用羟基磷灰石对漆酶进行了固定化研究,确定了羟基磷灰石对漆酶固定的最适条件,并研究了不同温度下游离漆酶与固定化漆酶活力的差异,同时也对游离漆酶与固定化漆酶的Km值进行了研究.结果表明:0.08 g羟基磷灰石固定1 mL粗酶液,当固定化过程pH为6,交联剂戊二醛浓度为6%,固定化时间为1 h时,酶活力达到最大.当温度在3...

Abstract:Immobilization of laccase to hydroxyapatite was researched,and the best condition of immobilization was confirmed.Otherwise,the distinction on the activity between immobilized laccase and dissociative laccase and the two laccases’Km were studied.The results showed that 1 mL original enzyme solution immobilization on 0.08 g hydroxyapatite,if the pH was 6.0,the concentration of glutaraldehyde was 6%,and the time of the immobilization was 1 h,the activity of the immobilized laccase would be the highest.The activity of the dissociative laccase would be increasing if the temperature ranges was 30~70 ℃.The activity of the immobilized laccase would be the highest when the temperature was 50 ℃.
