中空纤维超滤和疏水层析大规模纯化质粒DNA

摘    要:目的建立适合大规模纯化质粒DNA的工艺。方法采用碱裂解、中空纤维超滤浓缩、疏水层析、分子筛等分离技术纯化质粒DNA。结果纯化后的质粒DNA纯度达95%以上,整个工艺流程总产率为58%,各项指标均符合药用质粒DNA质量标准。结论此纯化工艺简便,产率较高,为DNA疫苗大规模纯化奠定了基础。

Abstract:Objective To develop a procedure for large-scale purification of plasmid DNA.Methods Purify plasmid DNA by a procedure consisting of alkaline lysis,ultrafiltration with hollow fiber filter,hydrophobic chromatography and molecular sieve chromatography.Results The purified plasmid DNA reached a purity of more than 95% and met the requirements for plasmid DNA for medical use.The yield of the procedure was 58%.Conclusion The developed purification procedure was simple and of high yield.It laid a foundation of large-scale purification of DNA vaccine.
