西藏淡红蜡伞菌多糖的提取及单糖组成研究

摘    要:采用正交试验优化西藏淡红蜡伞菌粗多糖提取工艺条件,通过蒽酮-硫酸法比色测定提取粗多糖中的总糖含量,并应用气相色谱法对淡红蜡伞菌多糖中单糖的组成进行分析。结果表明,西藏淡红蜡伞菌多糖最佳提取条件是提取时间4h,温度80℃,料水比1:10,乙醇沉淀浓度85%,多糖得率为6.52%;比色测定粗多糖中的多糖含量在49.17%~69.86%。色谱分析得出该多糖是由5种单糖组成的杂多糖。

Abstract:The extraction technological conditions of polyoses of the hygrophorus russula in Tibet were designed using orthogonal methods.The total sugar contents in the polyoses were assayed by anthrone- sulfuric acid colorimetric estimation,and the compositions of monosaccharide in polyoses which belonged to hygrophorus russula were analysed by employing Gas Phase Chromatography.The results showed as fol- lows:The optimum extraction time was 4h;the temperature was 80℃;the ratio of material and water was 1:10;the deposit concentration was 85%;the yield of polyoses was at the range of 49.17-69.86% by colori- metric estimation.The polyoses were composed by 5 monosaccharide using Gas Phase Chromatography.
