麦冬多糖的分离纯化及其组分的初步分析

摘    要:采用高温水提取低温乙醇沉淀的方法得到麦冬粗多糖,使用葡聚糖凝胶柱Sephadex G-100对已提取的麦冬粗多糖进行分离纯化。将纯化所得的麦冬多糖完全水解,分别以木糖、阿拉伯糖、果糖、甘露糖、葡萄糖、半乳糖等单糖色谱纯为标准品,采用薄层层析法和高效液相色谱法对上述全水解产物进行比对分析。TLC和HPLC定性分析发现麦冬多糖中单糖组成主要是果糖和葡萄糖,以果糖和葡萄糖的质量浓度与HPLC峰面积的线性关系定量分析出其组分的含量。结果表明,浙江慈溪产麦冬中多糖的糖单元基本组成是果糖和葡萄糖,并确定了其摩尔比为12∶1。

Abstract:Ophiopogon japonicus crude polysaccharides were obtained by water extraction and ethanol precipitation,and purified by Sephadex G-100.The hydrolytic products were analyzed by TLC chromatography and HPLC.Results showed that Ophiopogon japonicus polysaccharides were mainly composed of fructose and glucose.Basic compositions of sugar moiety of Ophiopogon japonicus polysaccharides from Zhejiang Cixi were Fru and Glc,and their molar ratio was approximately 12∶1.
