魔芋葡甘低聚糖硫酸酯抗柯萨奇病毒B3药理作用

用不同浓度HOGS的维持液培养Hela细胞,MTT法测定药物对细胞的毒性作用;培养上清液滴定TCID50;按HOGS与病毒的不同作用方式进行分组,采用CPE观察法结合MTT法得出其抗CVB3活性最好的一组并进一步得出其抗CVB3半数有效浓度及治疗指数.结果显示HOGS对细胞无明显毒性作用,半数中毒浓度TC50为0.167 7 mg/mL;HOGS对CVB3直接作用的效果随着时间的延长有所增强,中和3 h后效果最好,其半数有效浓度IC50为0.775 7 mg/mL治疗指数TI为4.63;HOGS对CVB3侵入细胞的阻断作用组随着时间的延长其效果也较有所增强,作用4 h后IC50为0.351 7 mg/mL的TI为2.10.通过对HOGS抗病毒药理作用的研究可证实HOGS具有一定的直接抗CVB3作用并且对细胞有较好的保护作用,对CVB3的吸附、穿入等环节随着时间的延长其效果较有所增强.

Study of the Inhibitory Effect on Coxsackievirus Group B-type 3 Replication in Vitro by Hydroxylpropyl Oligo-konjac Glucomannannuroate Sulfate

Hela cells were cultured with different concentration of HOGS and MTT colorimetric assay was used to get virus inhibitory rate;50% tissue culture infection dose(TCID50) of culture supernatant.There were four groups set up according to different effects on Hela cells and CVB3 by using cell infection model.The results of the cells were judged comprehensively by means of the observation with microscope combined with the detection with MTT.HOGS do not have obviously toxic effect on cells in vitro and the median toxic concentration(TC50) was 0.1677 mg/mL;HOGS showed the ability of directly-killing-virus and the median inhibitory concentration(IC50) of three hours was 0.7757 mg/mL,so the treatment index(TI) was 4.63;HOGS also showed the ability of adsorption blocking and the IC50 of four hours was 0.3517 mg/mL,so the TI was 2.1.The study goes to show that HOGS may kill viruses directly and protect cell group,the effect of preventing CVB3 from viral adsorption and penetration can reinforce as time lengthening.
