一种盐酸吡格列酮简便合成工艺研究

摘    要:报道了一种新的合成治疗糖尿病药物盐酸吡格列酮工艺。以活性KOH为缩合催化剂,5-乙基-2-羟乙基吡啶与对氟苯甲醛缩合得到4-{2-(5-乙基-2-吡啶基)乙氧基}苯甲醛,再与2,4-噻唑烷二酮进行缩合,利用复合型Ni-Cu-Fe催化剂催化加氢还原,再经盐酸酸化成盐得盐酸吡格列酮。

Abstract:To report a new synthesis process of pioglitazone hydrochloride for treating diabetes.With activated potassium hydroxide as catalyst,2-(5-ethyl-2-pyridyl)ethanol was condensed with 4-fluorobenzaldehyde to give 4-[2-(5-ethyl-2-pyridyl)oxethyl]benzaldehyde,then was condensed with 2,4-thiazolidinedione.The product pioglitazone hydrochloride was obtained by hydrogen reduction with composite catalyst of Ni-Cu-Fe,and then acidifying with hydrochloride acid.
