摘    要:研究了马蔺子甲素(Q)及其衍生物(Q1-Q8)对荷S180肉瘤小鼠的辐射增敏作用。应用动脉夹阻断局部血流以造成肉瘤乏氧的方法来研究对小鼠S180肉瘤乏氧细胞的辐射增敏作用。实验结果表明:与对照组相比,照射前腹腔注射Qn后,肉瘤氧增比减小,肉瘤体积变小,增敏比及肉瘤抑瘤率增加,表现出Qn对肉瘤的抑制作用。有氧和乏氧条件相比,乏氧条件的辐射增敏作用更明显。体外乏氧细胞毒比率的结果也表明,此类药物对乏氧

Abstract:The purpose of the paper is to investigate whether Irisquinone(Q) and its derivative(Q1-8) have heavy ion radiosensitization effect on 5180 sarcoma. Under aerobic and hypoxic condition, mice with 5180 were randomly divided into sir groups: control; r irradiation, C~6+ irradiation alone; C~6+ irradiation, r radiation +ip Qn; ip Q. alone. Hypoxic condition of tumor was caused by clamping the leg bearing tumor for 20 minutes. Qn was diluted with Tween-80 and saline and injected into the mice before irradiation. The dose of r irradiation and C~6+ irradiation was 18Gy and 6Gy, respectively. The volume of mouse's tumors was measured every two days. We calculated tumor growth rate, sensitzation enhancement ratio (SER), oxygen enhancement ratio (OEP). SER of Q, Q_7 and Q_8 with C~6+ irradiation on hypoxic tumor was larger than that on aerobic tumor. The OER decreased and the tumor inhibittion rate, hypoxiaselective cytotoxicity in vitro increased. For both aerobic and hypoxic conditions, the inhibition effect on tumors was more significant in hypoxic. There was no obvious radiosensitivity in aerobic condition r irradiation +Qn and C~6+ irradiation +Qn. It is suggested that Quinones could selectively increase the radiosensitivity of hypoxic tumor.
