电纺法制备酮洛芬酯类前体药物载药纤维

摘    要:以酮洛芬甲酯为模型药物,醋酸纤维素为载体高分子材料,采用传统静电纺丝法制备酮洛芬甲酯/醋酸纤维素载药纤维。通过SEM、FTIR对载药纤维的结构和形貌进行了研究。FTIR图谱显示酮洛芬甲酯已经成功载入醋酸纤维素纤维中,并且仍然保持原有的药物活性。对纺丝过程中的纺丝液流速和施加电压的影响进行考察,发现随着施加电压的增加,载药纤维能得到较好的形貌和直径,而且最佳纺丝液流速为0.8 mL/h;另外,通过对载药纤维的释药性能考察发现,载药纤维能较好地实现酮洛芬甲酯的缓控释,为药物经皮吸收研究提供了理论基础。

Abstract:Cellulose acetate(CA)fibers loaded with ketoprofen methyl ester were prepared through electrospinning.The chemical and morphological characterizations of the drug-loaded fibers were investigated by means of SEM and FTIR,as well as the studies of the drug release properties.The results from FTIR indicated that ketoprofen methyl ester was successfully encapsulated inside the CA fibers,and its pharmaceutic activity was sustained after the electrospinning process.The morphology and diameter of the fibers were influenced by the flow rate of spinning solution and applied voltage.It was found that the fibers with finer and uniform morphology and structure were obtained with increasing applied voltage,and the suitable flow rate of spinning solution was selected at 0.8 mL/h for further electrospinning process.Furthermore,from in vitro release drug-loaded fibers exhibited the sustained and controlled release behavior for ketoprofen methyl ester,and the present study could provide a basis for practical applications in transdermal drug delivery systems.
