摘    要:采用免疫亲和柱净化高效液相色谱法检测稻米中的黄曲霉毒素B1(AFB1)。稻米样品经甲醇/水(80/20, V/V)粗提取,再经免疫亲和柱净化和三氟乙酸衍生化,最后经高效液相色谱法测定。经检测,AFB1含量在0.01~2.50 ng范围内,所得到的AFB1标准曲线呈良好的线性关系,回归方程为Y=60.355x +0.054 3,相关系数为0.999 7。该方法检出限为0.1 μg/kg,加标回收率为89.26%~99.45%,SD为2.48%~3.74%。由于该方法操作的选择性高、灵敏度高、检测效果好,因此适于较大批量粮油作物中黄曲霉毒素的检测,便于示范推广。

Abstract:This study investigated the effect of high performance liquid chromatography ( HPLC ) with AflaTest affinity column on determination of aflatoxin B1 in rice. Rice samples were extracted with methanol/water (80/20, V/V) , followed by AflaTest affinity colmnn for cleanup and then trifluoroacetic acid for derivatization, finally detected by the method of HPLC-FLD. The curve of calibra- tion was well linear in the range of 0.01 -2.5 ng, Y = 60. 355 x + 0. 054 3, R^2 = 0. 999 7. Furthermore, the detection limit was 0.1 μg/kg, and the recovery of the spiked AFB1 ranged fi'om 89.26% to 99.45% , and the standard deviation ranged from 2.48% to 3.74%. Since the method had advantage of high selectivity, high sensitivity, and good detection effect, it was suitable for detecting the aflatoxin in massive grain or oil crops, which was favorable for demonstration and extension.
