H2O2对DNA影响的拉曼光谱分析

摘    要:检测了鲱精DNA水溶液及其经过1%、2%和3%H2O2处理48h的拉曼光谱图.实验结果表明DNA在水溶液中同时具有A、B两种构象,但以B型构象为主.经H2O2处理后,DNA水溶液的A型构象有所增强,碱基堆积程度下降.H2O2对碱基和脱氧核糖几乎没有损伤,对DNA的主链结构也只造成很轻微的损伤.由此可见H2O2本身对DNA的损伤是很小的,只有在外界因素诱发下H2O2经反应生成HO.才会给DNA造成严重损伤.

Abstract:The Raman spectra of DNA in aqueous solution treated by 1wt.-%,2wt.-% and 3wt.-% H_2O_2for 48h are measured.The Raman spectral measurements indicate that DNA in aqueous solution shows both A-type and B-type conformation with B-type conformation in the majority.A-type conformation of DNA in aqueous solution is dominated after treatment of H_2O_2 for 48h.H_2O_2 leads to decrease in the base stacking.H_2O_2doesn't damage the bases nearly,and only damages the backbone of DNA slightly.The damage of DNA is mainly from HO resulted from chemical reaction instead of H_2O_2 itself.
