血浆中柚皮素、橙皮素缀合物酸水解条件优化

摘    要:建立了大鼠血浆中柚皮素、橙皮素缀合物水解为游离柚皮素、橙皮素的酸水解方法,主要影响因素是盐酸浓度、水解温度、水解时间;通过单因素影响实验和正交实验相结合的方法进行优化。最终确定的水解方法是含药大鼠血浆加入等体积4 mol·L-1的盐酸在70℃下水浴加热1 h。此方法对目标产物柚皮素、橙皮素的破坏较小,可应用于血浆样本中总柚皮素、总橙皮素的含量测定。

Abstract:The acid hydrolysis method was established to convert naringenin and hesperetin conjugates to free naringenin and hesperetin in rat plasma. The main affecting factors were the concentration of hydrochloric acid, hydrolysis temperature, and hydrolysis time. The final hydrolysis conditions were obtained through orthogonal experiments as well as single factor affecting experiments. The final hydrolysis process was adding an equal volume of 4 mol . L^-1 hydrochloric acid to rat plasma after drug administration and then the mixture was heated at 70 ℃ water bath for 0. 5 h. This method can be applied to the determination of total naringenin and total hesperetin determination in rat plasma with a small degree of destruction of the target compounds.
