亳菊不同部位绿原酸及总黄酮的含量测定

摘    要:研究亳菊不同部位绿原酸及总黄酮的含量。绿原酸采用Phenomenex C18色谱柱(250 mm×4.6 mm,5μm),乙腈-0.4%磷酸水溶液(12∶88)洗脱,检测波长为327 nm;总黄酮采用比色法进行测定,以亚硝酸钠-硝酸铝为显色剂进行测定,检测波长为510 nm。结果表明:亳菊不同部位绿原酸的含量分别是根(0.39%)、茎(0.42%)、叶(0.72%)、花(0.54%);总黄酮的含量分别是根(10.30%)、茎(9.24%)、叶(35.69%)、花(18.08%)。亳菊叶中绿原酸及总黄酮的含量最高。

Abstract:The content determination in different parts of Chrysanthemum morifolium Ramat was studied.The content of chlorogenic acid was determined by HPLC on Phenomenex C18 (250 mm ×4.6 mm, 5 μm) column with CH3 CN-0.4%H3 PO4 ( 12:88 ) as mobile phase.The absorbance was monitored at 327 nm.The content of flavonoid was determined by spectrophotometry, with NaNO2 -Al( NO3 ) 3 as chromogenic agent.The absorbance was monitored at 510 nm.The results showed that the different parts of chlorogenic acid in Chrysanthemum morifolium Ramat was different: root(0.39%) , steam(0.42%) , leave(0.72%) , and flower(0.54%).The different parts of flavonoid was different: root ( 10.30%) , steam ( 9.24%) , leave ( 35.69%) , and flower ( 18.08%).The highest level of Chlorogenic acid and flavonoid was in leaves.
