玉米芯木聚糖的酶法降解及产物的分离纯化

摘    要:以自制玉米芯木聚糖为原料,采用单一及复合木聚糖酶处理,研究其对降解产物产率的影响,比较了不同的分离纯化方式对降解产物得率的影响,并采用HPAEC图谱分析产物纯度。研究结果表明,单一木聚糖酶降解能力较差,而在两种木聚糖酶共同作用下的低聚木糖得率不仅能增加木二糖和木三糖的产率,也能大幅减少一般的反应时间。活性炭与硅藻土法批式分离木聚糖酶解液总回收率约在20%左右,并且木二糖较多出现在活性炭CJ组5%以下的EtOH冲提液内;将1.0%玉米芯木聚糖酶水解液依序通过不同分子量切,最后经1kDa薄膜过滤后可收得聚合度7以下的低聚木糖产物11.5% ̄15.2%;活性炭!硅藻土混合管柱连续分离低聚木糖,活性炭CJ组5%乙醇冲提管柱可获得13%的木二糖,经HPAEC图谱分析连续式分离能得到纯度较高的单一低聚木糖划分物,但是其回收率不及批式分离的高。

Enzymolysis of corn cob xylan and separation of degradation products

Abstract:With self-made corn cob xylan as the material,the influences of one or compounded xylanases and different separation and purification methods on the yield of degradation products were studied and compared,and their purity analyzed by the HPAEC atlas. The results indicated that one xylanase had a poor degradation effect,but two xylanases used together not only resulted in an increased yield of xylobiose and xylotriose,but also greatly shortened the reaction time. The recovery of xylo-oligosaccharide,which was from xylan hydrolyzed by xylanase and separated by activated carbon and celite in a batch mode,was about 20% and X2 principally appeared in the extracted solution which contained 5% ethyl alcohol in CJ group. 1% solution,which had been hydrolyzed by xylanase,was passed through different molecular weight cutoff in turn and finally filtrated by 1kDa membrane,there was a recovery of 11.5% - 15.2% for xylo-oligosaccharide which had a degree of polymerization under 7; extracted solution,which was separated by an activated carbon-celite pipe string in a continuous mode and involved 5% ethyl alcohol in CJ group,contained 13% X2,and after analyzed by the HPAEC atlas,there was a result that separation in a continuous mode would get more highly purified products but have lower recovery than separation in a batch mode.
