摘    要:采用自行设计的RT-PCR试剂盒。在自制的由聚合酶链反应(PCR)——毛管电泳(CE)芯片、芯片热循环仪和激光诱导荧光芯片分析仪组成的微流控芯片系统上,对SARS病毒(重症急性呼吸综合症)和18例SARS患者咽拭子样品连行了分析和检测,实现了聚合酶链反应和电泳检测的微流控芯片在线分析。微流控芯片系统具有高检潮灵敏度、高分辨率、高检出率等优点,试剂消耗少和检测时间短,可能成为SARS早期检测的一种新的手段。

Abstract:The microfluidic chip system, which includes microfabricated electrophoretic devices, chip thermal cycler, laser confocal fluorescence analyzer and the related reagents, has been used for the anti-SARS issue. The genomic fragments of severe acute respiratory syndrome associated coronavirus (SARS-Cor) and 18 specimens of respiratory secretions of patients with SARS were amplified by the chip-PCR, and then the product of chip-PCR was on-line detected by the microfluidic analyzer. The results show that the microfluidic chip system has the advantages of high detection limit, high resolution rate, high sensitivity, low reagent consumption and fast detection.
