富锌酵母的选育及其培养工艺

摘    要:以紫外谤变配合驯化的方法获得富锌酵母HB524,并在单因素研究的基础上通过正交实验确定了该菌株富锌培养的最佳条件为:麦芽汁4Bx,葡萄糖25g/L,尿素1.0g/L,磷酸二氢钾0.5g/L,不加豆芽汁,接种体积分数10%,6mmol/L锌在接种后4h内加入,48h摇瓶培养后富锌总量可达到130mg/L。球磨法破壁厦锌的浸出结果表明,至少47.84%的锌是有机态形式结合的,其余锌松散结合在细胞壁或游离于细胞内部。

Abstract:A zinc-enriched yeast zinc-enriched yeast strain,namely HB54,was isolated from UV mutation and domestication.Then the nutritional and environmental conditions were optimized with single factor experiment and orthogonal design.A total zinc of 130 mg/L was achieved at 48 h in flask culture with the optimum conditions: 4 Bx malt,25 g/L glucose,1.0 g/L urea,0.5 g/L KH_2PO_4,10% inoculum size and 6 mmol/L Zn~(2+) was added at 4 h after inoculation.The results from the mechanical ball milled yeast cells and the leaching of zinc illustrated that more 47.84% zinc in organic and the rest zinc was adhered to the cell wall or dissociated in the cell.
