铁皮石斛无菌萌发及小苗快繁培养条件研究

摘    要:以铁皮石斛成熟未开裂果实中的种子为外植体,比较了不同培养基配方中种子的萌发情况以及小苗增殖情况,通过组织快繁的方式获得了铁皮石斛壮苗.研究表明,铁皮石斛种子无菌发芽的最适培养基为WPM培养基,增殖最适培养基为1/3MS+香蕉泥(200 g/L)+蔗糖(30 g/L),快速繁殖过程无需添加激素.组培快繁技术的成功为铁皮石斛种苗的工厂化生产奠定了技术基础.

Abstract:The seeds from mature but not dehiscent fruit of Dendrobium officinale Kimura et Migo were chosen as explant for aseptic germination.The germinating state and plantlet proliferation in different culture medium were investigated and well grown seedlings were got.The results show that the optimal medium for seed aseptic germination of Dendrobium officinale Kimura et Migo is WPM,while the optimal medium for proliferation is 1/3MS+200 g/L of Banana puree+30 g/L of sucrose without any plant hormone.The results lay the foundation of industrial plantlet reproduction of dendrobium officinale Kimura et Migo.
