巴里坤草原西伯利亚蝗虫肠道内高产纤维素酶菌株筛选

摘    要:从采集到的新疆巴里坤草原西伯利亚蝗虫活体标本肠道内分离筛选高产纤维素酶细菌菌株并对其进行初步鉴定。采用常规方法分离肠道菌;利用羧甲基纤维素钠刚果红(CMC-Na)改良培养基初筛分解纤维素菌株,选取透明水解圈较大的菌株测其纤维素酶活力进行复筛;利用VITEK-32生化鉴定仪(法国梅里埃公司)对复筛菌进行鉴定。实验结果表明以羧甲基纤维素钠刚果红(CMC-Na)培养基作为初筛培养基效果较好,初筛到30株菌,选取水解圈较大的10株菌进行复筛,复筛得到4株产纤维素酶活性较高的菌株。经初步鉴定其中一株菌可能是干燥棒杆菌;另一株菌可能是耳葡萄球菌,两株菌未鉴定出。

Abstract:The high-yield cellulase strains were found from the intestinal of Gomphocerus Ssibiricus.The intestinal bacteria were isolated through the conventional methods.They were preliminarily identified by using cellulose-Congo red medium,secondly screened according to their cellulase activities by hydrolysis circle size and finally identified by VITEK-32.The results indicated that cellulose-Congo red medium is a better screening medium.By it 30 bacteria were isolated.4 high-yield cellulase strains were screened from 10 strains with larger hydrolysis circle size.Among them Corynebacterium xerosisthe and Staphylococcus auricularis were identified,but 2 other strains couldn't be identified.
