流感嗜血杆菌多糖抗原主要成分的分析

摘    要:按Schneerson方法提取了2批多糖抗原,每批包括a、b、c、d、e、f6个血清型,并测定了抗原的固总、蛋白、核酸、磷、核糖、葡萄糖及氨基己糖。并用纸层析作进一步鉴定,用反向血凝方法测定血清学特性。结果表明,HK658(c)主要成分为氨基葡萄糖,HK646(f)为氨基半乳糖,HK644(d)为氨基己糖,HK645(a)为氨基己糖及葡萄糖,HK179(b)、HK641(e)主要为核糖,HK641并含有氨基己糖。6个血清型多糖均是型特异抗原,与相应型的IgG致敏血球能产生凝集,且最高滴度可达1:51200。

Abstract:2 lots of Hemophilus influenzae polysaccharide antigens (including types a, b, c,d, e and f) were extracted according to Schneerson's methed and examined for total solid, protein, nucleic acid, phosphorus, ribose, glucose and aminohexose contents, then further characterized by paper chromatography. The serological characteristics of the 2 lots were examined by reverse hemagglutination. The result showed that the essential components of HK658 (c), HK646(f), HK644 (d), HK645 (a), HKl79 (b) and HK641 (e) were aminoglucose, galactosamine, aminohexose, aminohexose and glucose, ribose and ribose respectively. HK641 also contained aminohexose. All the polysaccharides of 6 serotypes were type specific antigens which could agglutinate with corresponding IgG sensitized blood cells, and the highest titer of them reached 1:51200.
