中国长春地区汉族人MMP-9C-1562T基因多态性与IgA肾病易感性的相关性

摘    要:目的探讨中国长春地区汉族人基质金属蛋白酶-9(MMP-9)C-1562T基因多态性与IgA肾病易感性的相关性。方法应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)分别检测长春地区汉族88例IgA肾病患者和133例健康对照者的MMP-9C-1562T基因多态性,比较两组的基因型和等位基因的频率,分析其与IgA肾病易感性的相关性。结果IgA肾病患者MMP-9C-1562T基因型频率分布为:CC型65例(0.739),CT型23例(0.261),TT型无;健康人的频率分别为:CC型77例(0.579),CT型53例(0.398),TT型3例(0.023),两组基因频率分布差异具有统计学意义。IgA肾病患者和健康人等位基因C频率分别为0.869和0.778,等位基因T频率分别为0.131和0.222,两组等位基因的分布频率差异也具有统计学意义。结论中国长春地区汉族人MMP-9C-1562T基因多态性可能与IgA肾病的易感性有关,C等位基因可能是其易感基因。

Relationship between Matrixmetalloproteinase-9(MMP-9)C-1562T Gene Polymorphism and Susceptibility to IgA Nephropathy in Hans in Changchun Region,China
Abstract:Objective To investigate the relationship between matrixmetalloproteinase-9(MMP-9)C-1562T gene polymorphism and susceptibility to IgA nephropathy in Hans in Changchun Region, China. Methods The MMP-9 C-1562T gene polymorphisms of 88 patients with IgA nephropathy and 133 healthy persons were tested by PCR-RFLP, based on which the genotypes and allelic gene frequencies of the two groups were compared to analyze the relationship to susceptibility to IgA nephropathy. Results Of the 88 patients with IgA nephropathy, 65 (0. 739)and 23 (0. 261)were of genotypes CC and CT respectively, while none were of genotype TT. However, of the 133 healthy persons, 77(0. 579), 53(0. 398)and 3(0. 023)were of genotypes CC, CT ad TT respectively. Significant differences were observed between the gene frequency distributions of the two groups. The frequencies of allelic gene C of patients with IgA nephropathy and healthy persons were 0. 869 and 0. 778, while those of allelic gene T were 0. 131 and 0. 222, respectively, which showed significant difference. Conclusion The MMP-9 C-1562T gene polymorphism in Hans in Changchun Region might be related to the susceptibility to IgA nephropathy, and allelic gene C might be a susceptibility gene.
