摘    要:本文报道链霉亲和素(streptavidin.SA)的产生、纯化和主要特性鉴定。根据Streptomycesavidinii菌对碳、氮源的需求,配制成特定培养基,在28℃±1℃摇床培养S。avidinii 10天,SA产量为10~50mg/L。用2-亚氨基生物素琼脂糖凝胶层析纯化,每升培养上清平均可提取SA20~30mg。所得SA电泳出现2~3条区带,分子量为64562,等电点为6.0,紫外吸收扫描图谱与Sigma产品相似,SA活性平均为14.4 μg/ml,表明自制SA可供实际应用。

Abstract:The production, purification and Characterization of streptavidin(SA) were described in this report. Based on the comparative study of requirements of carbon and nitrogen sources by S. avidinii, the special medium was designed and prepared by shaking culture at 28℃±1℃ for 10 days. The productivity of SA was 10~50 mg/L. After purification by 2-imminobiotin agarose gel chromatography, 20~30 mg of SA could be obtained from 1 L of culture supernant. 2~3 bands were appeared on the electrophoresis of SA. The molecular weight of SA was 64652 with PI 6.0. The spectrum scanning of ultraviolet absorption was similar with the same product of Sigma Company. The averaged activity of SA produced was 14.4μg/ml. The results showed that the SA produced by authors could be provided for immunological applications.
