摘    要:甘氨酸和琥珀酸是红假单胞菌生物转化合成5-氨基乙酰丙酸(ALA)的前体。在异养培养的条件下考察了两者浓度,添加时间以及协同作用对红假单胞菌生长及累积ALA的影响。结果表明添加100mmol/L甘氨酸,使ALA比生成速率(qALA)明显提高,ALA产量达到22.2mg/L,是不添加甘氨酸的3倍;同时加入100mmol/L的甘氨酸和20mmol/L的琥珀酸,ALA产量达到29.4mg/L,是两者都不加时的4.1倍。

Abstract:Glycine and succinyl-CoA are precursors of 5-aminolevulinic acid(ALA) biosynthesis. The effects of the addition concentration of glycine and succinate, as well as both glycine and succinate mixture and different additional time of glycine on the growth of Rhodopseudomonas sp. and ALA production in shake flask were investigated. The experimental results showed that specific ALA production rate(q_(ALA)) was enhanced obviously with the addition of 100mmol/L glycine into the medium, and the production of ALA reached 22.2mg/L, which is 3 times of that in the culture without glycine addition. It was also found that the addition of 100mmol/L and 20mmol/L succinate mixture to the medium was able to increase the ALA productivity up to 29.4 mg/L, or 4.1 times higher than that of control.
