双水相系统分离纯化南极假丝酵母脂肪酶

摘    要:对南极假丝酵母脂肪酶(CAL)在双水相中的分配情况进行了研究,考察了温度、聚乙二醇(PEG)相对分子质量、NaC l质量分数和(NH4)2SO4质量分数对分配系数K(CAL)的影响,结果表明:温度对K(CAL)影响不大;低相对分子质量PEG与蛋白质的疏水作用可促进CAL的分配;二相间电势差等对分配平衡、有较大影响。在PEG2 000/(NH4)2SO4双水相体系中,CAL最佳的萃取分离条件为:室温下,质量分数20%PEG2 000,12%(NH4)2SO4,1.5%NaC l,此时,K(CAL)为8.16,CAL回收率为91.2%。

Abstract:The partition behavior of Candida antarctica lipase(CAL) in aqueous two-phase system(ATPS) was investigated.The effects of temperature,relative molecular mass of polyethylene glycol(PEG),the mass fraction of NaCl and(NH4)2SO4 on partition coefficient of CAL K(CAL) were studied.The results show that in ATPS,temperature was unimportant for partition coefficient of CAL K(CAL),while the electric potential difference between top and bottom phase was important.It was also discovered that the less the molecule mas...
