| 摘 要： | 目的建立高效液相色谱法(HPLC)测定八正片中大黄素及大黄酚含量。方法采用菲罗门C18柱(4.6 mm×250mm,5μm);流动相:乙腈-甲醇-0.1%磷酸溶液(42:23:35);流速:1.0 mL/min;检测波长:254 nm;柱温:30℃。结果大黄素、大黄酚的线性范围分别为0.0267~0.4272μg、0.0417~0.6677μg,总大黄素、总大黄酚、游离大黄素、游离大黄酚的平均加样回收率分别为97.88%(RSD=1.2%)、97.22%(RSD=1.1%)、96.09%(RSD=0.7%)、96.70%(RSD=0.9%)。结论本方法准确、可靠,可用于八正片中大黄素、大黄酚的含量测定。

| Abstract: | Objective To establish a method for determination of emodin and chrysophanol in Bazheng Tablet by HPLC.Methods The separation was performed on Phenomenex C18 column（4.6 mm× 250 mm,5 μm）.The mobile phase was composed of acetonitrile-methanol-0.1％phosphoric acid（42∶23∶35）; the column temperature was set at 30 ℃; the flow was 1.0 mL/min; the wavelength of detection was 254 nm.Results The calibration curves showed good linearity in the range of 0.0267-0.4272 μg（emodin） and 0.0417-0.6677 μg（chrysophanol）.The average recoveries were 97.88 ％（RSD=1.2％）,97.22 ％（RSD=1.1％）,96.09％（RSD=0.7％） and 96.70 ％ （RSD=0.9 ％）,respectively.Conclusion The method is accurate and reproducible.It can be used for the determination of emodin and chrysophanol in Bazheng Tablets. |
