羊骨酶解物中降血压肽的分离、纯化工艺研究

对不同pH和温度等条件下的羊骨酶解物新中7号和中14号进行SephadexG-25层析，并对层析后的收集液冷冻干燥，利用Tricine-SDS-PAGE电泳检测其分子量。结果表明：（1）新中7号和中14号羊骨酶解物，酶解8h的ACE活性最高；（2）新中7号羊骨酶解物酶解8h分离出来的蛋白质谱带的分子量在20100U以上，酶解10h分离出来的蛋白质谱带的分子量在7823U以上；（3）中14号羊骨酶解物酶解6h分离出来的蛋白质谱带的分子量14400U以上，酶解8h分离出来的蛋白质谱带的分子量在20100U以上。

SEPARATION AND PURIFICATION CRAFT RESEARCH OF FALLING THE BLOOD PRESSURE PEPTIDE IN THE SHEEP BONE ENZYMOLYSIS

To different condition under pH and temperature and so on sheep bone enzymolysis new 7 and 14 carry on the sephadexG-25 chromatographic analysis. After chromatographic analysis drips freeze-drying, examines its molecular weight by using the Tricine-SDS-PAGE electrophoresis. The result indicated that:(1)for the sheep bone enzymolysis new 7 and 14,the enzymolysis for 8 h ACE activeness is the highest;(2)the sheep bone enzymolysis new 7 the enzymolysis 8 h , separation protein band molecular weight above 20 100 u , and the sheep bone enzymolysis new 7 the enzymolysis 10 h , separation protein band molecular weight above7 823 u;(3)the sheep bone enzymolysis 14 the enzymolysis for 6 h, separates protein band molecular weight above 14 400 u, and the sheep bone enzymolysis 14 the enzymolysis for 8 h, separates protein band molecular weight above20 100 u.
