融合蛋白PTD-SOD对紫外辐照下L-02细胞的保护和修复能力的研究

摘    要:通过体外细胞培养,探讨了融合蛋白PTD-SOD是否径由跨膜转导而增强SOD对靶细胞紫外辐射损伤防护和修复能力.结果表明辐照时间为120s,酶活力为320U/mL时,PTD-SOD对细胞的保护率达58.5%,约为野生型SOD的5倍;紫外辐照时间为140s,酶活力为500U/mL时,PTD-SOD对细胞的保护率仍有43.2%,相同情况下野生型SOD的保护率趋近于0;辐照时间40s,酶活力为500U/mL时PTD-SOD对细胞的修复率有50.1%,而野生型SOD的修复率最高也没有超过8%.这些结果说明相对野生型SOD,PTD-SOD对L-02细胞的保护和修复能力得到了显著的增强,能有效地提高SOD的生物利用率.

Abstract:Cell culture was carried out to investigate whether the protective and repairing capacity of SOD against the radiation damage induced by UV-C was improved because of its transduction ability. When L-02 cell line being exposed to UV-C in the solution with SOD enzymatic activity of 320U/mL, the protective rate of PTD-SOD increased to 58.5%, which was about 5 times of that of wild-type SOD; when irradiation duration prolonged to 140s, the protective rate of PTD-SOD remained to be 43.2%, while that of wild-type SOD was almost undetectable. Regarding restorative rate, that of PTD-SOD could reserve 50.1% when L-02 cell exposed to UV-C for 40s, while that of wild-type SOD was less than 8%. These results suggested PTD enhanced the protective and repairing ability of SOD against UV radiation remarkably, owing to improve bioavailability of SOD.
