玛咖生物碱通过调节小鼠免疫力抑制结肠癌细胞的成瘤能力

摘    要:研究观察玛咖生物碱对小鼠结肠癌CT-26细胞成瘤能力的影响并初步探讨其可能机制。构建BALB/c小鼠CT-26细胞移植瘤动物模型,随机分为玛咖生物碱高剂量组、玛咖生物碱低剂量组及生理盐水模型对照组。连续灌胃用药27 d,绘制小鼠体重增长曲线及移植瘤体积增长曲线。第28 d处死小鼠,计算抑瘤率、相对肿瘤增殖率、脾脏指数;ELISA法检测血清IL-2、IFN-γ水平;HE切片观察移植瘤、脾脏、肝脏及肾脏组织学形态变化。结果表明,与对照组比较,实验组移植瘤重量明显降低(p<0.05),抑瘤率分别为43.37%和45.26%,组织学观察实验组移植瘤坏死面积较对照组明显增大。实验组小鼠脾脏指数明显高于对照组(p<0.05),且实验组脾脏白髓增生较显著。同时,血清IL-2及IFN-γ水平均明显高于对照组(p<0.05)。可见,玛咖生物碱可以通过提高小鼠免疫力抑制CT-26细胞的成瘤。
