阿司匹林-羟乙基纤维素复合物的合成

摘    要:以羟乙基纤维素和乙酰水杨酰氯为原料,合成了高分子复合物阿司匹林-羟乙基纤维素,用红外光谱仪对其结构进行了表征并用紫外分光光度计测定了阿司匹林的接入率。考察了缚酸剂、反应温度、配料比、反应时间等对反应的影响。结果表明,三乙胺作为反应的缚酸剂效果较好,吡啶和浓氢氧化钠水溶液则不合适;阿司匹林的接入率随反应温度以及阿司匹林与羟乙基纤维素的质量比的升高而增大;当选用DMF做溶剂、三乙胺(TEA)做缚酸剂、m(阿司匹林):m(羟乙基纤维素)=3.60∶1.32、温度60℃时反应6小时,所得复合物的接入率可达到31.18%。

Abstract:In order to obtain a sustained-release aspirin derivative in this paper,aspirin-ethylol cellulose coupling compound was synthesized by coupling reaction between ethylol cellulose and acetoxybenzoyl chloride.Its structure was characterized with FTIR and its drug-loading ratio was measured by ultraviolet spectrophotometer.Influences of various factors,such as acid absorbent,reaction temperature,mass ratio of reactants,reaction time were investigated in this paper.The results showed that triethylamine was most effective acid absorbent in this reaction,while pyridine and sodium hydroxide solution could not be used as acid absorbent.The drug-loading ratio of aspirin-ethylol cellulose coupling compound increaseed with the increases of reaction temperature and mass ratio of aspirin to ethylol cellulose.The reaction of acetoxybenzoyl chloride was carried out at 60 C for 6 hour when choosing DMF as reaction solvent and triethylamine as acid absorbent and the mass ratio of aspirin to ethylol cellulose was 3.60:1.32.Under these optimum reaction conditions,the drug-loading ratio 31.18% was attained.
