氟碳化合物FC-77——脂质体囊泡的制备及性能研究

摘    要:通过19F NMR、紫外可见光谱、激光粒度仪、Zeta电势和透射电子显微镜等方法研究了生理条件下(pH7.4)FC-77与脂质体囊泡的相互作用。结果表明,脂质体囊泡使得FC-77分子的紫外吸收光谱发生明显位移;FC-77使得脂质体囊泡的粒径和电势均增加。进一步的研究表明,FC-77能够引起囊泡双层膜通透性的降低。为开发FC-77的脂质体药物、增加FC-77的临床利用率提供了基础。

Abstract:The FC-77-loaded liposome vesicles are prepared and characterized using19F NMR,UV-vis spectrum,dynamic light scattering and transmission electron microscopy.Furthermore,the interaction mechanism between perfluoro-compound FC-77 and liposome vesicles are investigated.The results show that FC-77 molecule can be solubilized in liposome vesicle and the UV-vis absorption bands of FC-77 are obviously changed by the liposome vesicles.On the other hand,FC-77 molecules raise the diameter and zeta potential of liposome vesicles and lower the permeability of liposome vesicles.In a word,the perfluoro-compound FC-77 can interact with liposome vesicles and be loaded by liposome vesicles,which provide the base for developing FC-77-loaded liposome and increasing clinical utility.
