白藜芦醇磺酸酯的合成及抗氧化、抗肿瘤活性的研究

摘    要:以白藜芦醇和2, 4-二硝基苯磺酰氯为原料,通过酯化法得到白藜芦醇磺酸酯,通过NMR和LC-MS确定了该化合物结构。以维生素C为阳性对照,研究白藜芦醇磺酸酯对2, 2-联苯基-1-苦基肼(DPPH)自由基和2, 2′-联氮-双-3-乙基苯并噻唑啉-6-磺酸(ABTS)自由基的清除活性和抗氧化活性;噻唑蓝(MTT)法研究其对人肺癌细胞A549的增殖活性。实验结果表明,目标产物收率为33.1%。同时,目标产物在浓度为1.0 mg/mL时,对DPPH自由基和ABTS自由基的清除率分别为31.6%和42.7%,抗氧化活性均高于白藜芦醇,但低于维生素C;目标产物对人肺癌细胞A549具有较好抑制作用,IC50值为(10.2±0.37)μg/mL。

Abstract:The resveratrol sulfonate was obtained by esterification method using resveratrol and 2, 4-dinitrobenzenesulfonyl chloride as the starting materials, and the structure of the compound was confirmed by NMR and LC-MS. Using vitamin C as a positive control, study resveratrol sulfonate on 2, 2-biphenyl-1-picrylhydrazyl(DPPH) radical and 2, 2′-diazide-bis-3-B Scavenging activity and antioxidant activity of benzothiazoline-6-sulfonic acid(ABTS) radicals;The proliferation activity of human lung cancer cell A549 was studied by thiazolyl blue(MTT) assay. The experimental results show that the target product yield is 33.1%. At the same time, when the target product was 1.0 mg/mL, the scavenging rates of DPPH free radicals and ABTS free radicals were 31.6% and 42.7%, respectively, and the antioxidant activities were higher than resveratrol, but lower than vitamin C;The product has a good inhibitory effect on human lung cancer cell line A549, and the IC50value is(10.2±0.37) μg/mL.
